Difference between Dose, Dosage, Dosage form and Dosage Regimen:

1. Dose

- Definition: The specific amount or quantity of a medication administered at one time

- Examples:
- 500mg of Paracetamol
- 10mg of Amlodipine
- 20 units of Insulin

- Key Point: Dose refers to the exact amount of drug given at once

2. Dosage

- Definition: The amount of medication and the frequency of administration over a period

- Examples:
- 500mg twice daily
- 10mg once daily
- 20 units three times per day
- Key Point: Dosage includes both the amount and time component

3. Dosage Form

- Definition: The physical form in which a drug is produced and dispensed

- Examples:
- Tablets
- Capsules
- Liquid suspensions
- Injections
- Patches
- Unit dose packages (commonly used in long-term care facilities)

- Key Point: Different dosage forms are designed for different routes of administration and patient needs

4. Dosage Regimen

- Definition: The complete schedule of medication including:
- Dose amount
- Frequency of administration
- Duration of treatment
- Route of administration

- Examples:
- Amoxicillin 500mg capsules, taken three times daily for 7 days
- Metformin 850mg tablets, one tablet twice daily with meals, long-term use
- Prednisolone 5mg tablets, two tablets in the morning for 5 days, then one tablet for 5 days, then half tablet for 5 days

- Key Point: Dosage regimen is the complete therapeutic plan that healthcare providers prescribe

Real-World Example:
Let's take the example of Amoxicillin antibiotic treatment:

- Dose: 500mg (the amount given at once)
- Dosage: 500mg three times per day
- Dosage Form: Capsules
- Dosage Regimen: 500mg capsules taken three times daily for 7 days, to be taken with or without food.

Happy 71st Anniversary of Independence Day 9 November

Today is the day that Cambodia celebrates the 71st anniversary of the Independence Day on 9 November (9 November 1953-9 November 2024), when Cambodia gained its independence from French colonialism, coinciding with the 71st Anniversary of the establishment of the Cambodia Army.

Independence Day, November 9 reminded of the royal mission for the homeland and the Khmer people of His Majesty King Norodom Sihanouk, the father of independence, the King Ratanakoth, who gave up his strength and intellect to work hard in the framework of the royal duties to claim the true independence in Cambodia.

🔵Shelf-life of Medicinal Products (Expiration Period) as per ICH Q1A:

The time period during which a drug product is expected to remain within the approved shelf-life specification provided that it is stored under the conditions defined on the container label.

🔵How the shelf life is determined?
Drug applicants to provide stability testing data with a proposed expiration date and storage conditions when they apply for respective agency approval of their drug. This testing is designed to provide confidence that the product will meet the applicable standards of strength, quality, and purity throughout its shelf-life.
The expiration date/shelf-life date is calculated by considering the release of the batch if released within 30 days of the production.
The date of production of a batch is defined as the date that the first step is performed involving combining the active ingredient with other ingredients (excipients). For medicinal products consisting of a single active ingredient filled into a container, the initial date of the filling operation is taken as the date of production.

🔹 Zone I: Temperate Zone
🔹Zone II: Mediterranean/Subtropical zone
🔹Zone III: Hot dry zone
🔹Zone IVa: Hot humid/tropical zone
🔹Zone IVb: Hot/higher humidity

Stability testing is a critical process in the pharmaceutical industry used to determine how the quality of a drug substance or drug product varies with time under the influence of environmental factors such as temperature, humidity, and light. This testing helps ensure that the drug remains safe, effective, and of high quality throughout its shelf life.

As the environmental conditions are different around the world, accordingly regions are classified into various zones, as mentioned in my last post.
Here is a citation of a few territories per the assigned climatic zone for the stability testing.
