Advancing Sustainable Science: Circular Economy in Food Testing Laboratories 🌱🧪

At the AOAC SEA Section 5th Annual Conference, Ms. Aliefi Mutiara Syafitri from Kalbe Nutritionals Analytical Center (KNAC) presented a groundbreaking initiative: "Circular Reuse of Kjeldahl Distillation By-products for Sustainable Air Pollution Control in Food Testing Laboratory."

Protein testing—specifically through the Kjeldahl method—generates significant alkaline distillation waste. Rather than treating this strictly as hazardous waste (B3) needing disposal, the KNAC team designed a circular system that repurposes 2.5 tons of distillation by-products into scrubbing media for air pollution control wet scrubbers.

Key Impacts Achieved:

> 100% Reuse of distillation by-products as liquid wet-scrubber media.
> 156.8 kg/year reduction in raw chemical usage (NaOH).
> 45.8 Million IDR in annual cost savings.
> 100% Compliance with local air quality regulations.
> 49% Reduction in overall CO2 emissions per analysis from 2023 to 2025 through comprehensive Green Lab KPIs.

By aligning laboratory operations with the 12 Principles of Green Chemistry and Sustainable Development Goals (SDGs 9, 12, and 13), KNAC continues to prove that rigorous analytical quality and environmental sustainability can go hand-in-hand.

At the AOAC SEA Section 5th Annual Conference, Hendy Dwi Warmiko, S.Si., M.M. (General Manager of Corpora Science), presented pioneering research on identifying peptide markers to differentiate porcine and bovine gelatin in food products.

The Analytical Challenge:

Gelatin is widely used across the food, pharmaceutical, and cosmetic industries. However, distinguishing porcine from bovine gelatin presents significant analytical hurdles due to high sequence homology (over 95–97% similarity in Type I collagen chains) and the degradation of DNA during heavy industrial processing, which can limit traditional PCR testing in processed products like gummy candies and marshmallows.

Key Technological Insights:

> Optimized Sample Extraction: The presentation highlighted that Tris-HCl buffer (pH 8.0) outperforms standard ammonium bicarbonate for initial protein extraction in high-gel, sugar-rich matrices.

> Beyond Conventional Methods: While traditional LC-MS/MS (SRM/MRM) relies on pre-selected precursor-product ion pairs and specific collision energy optimizations, Parallel Reaction Monitoring via LC-Orbitrap High-Resolution Mass Spectrometry (PRM-LC-HRMS) simultaneously captures all product ion transitions at high mass accuracy.

> Enhanced Accuracy: This high-resolution approach enables full amino acid sequencing and precise identification of hydroxyproline modifications, significantly reducing the risk of false positives or false negatives in halal verification.

Corpora Science continues to support rigorous, data-driven methodologies to ensure authenticity and consumer trust in halal food products.

At the AOAC SEA Section 5th Annual Conference held at the Hotel Borobudur Jakarta, Jakarta, Indonesia, Heryani from the LPPOM MUI Laboratory presented a proposal for a Standard Method Performance Requirement (SMPR®) tailored for qualitative species-specific DNA detection using real-time PCR (qPCR).

As Indonesia prepares for expanded mandatory halal implementation under Law No. 33/2014 and PP No. 42/2024, laboratory testing plays a vital role in providing clear, analytical evidence for halal assurance. However, testing across different laboratories often suffers from variation due to differences in primers, targets, extraction efficiency, and statistical interpretation rules.

The presentation addressed this challenge by outlining a standardized performance framework. Key takeaways from the proposed draft include:

> Defining the Performance Baseline: Establishing precise probability of detection parameters (POD >/= 0.95, with an LOD 95% floor between 2.996 and 20 target DNA copies per PCR reaction).

> Rigorous Specificity & Controls: Requiring both in-silico alignment and experimental inclusivity/exclusivity challenges to prevent false positives and false negatives.

> Matrix-Level Verification: Ensuring validation covers the full analytical process—from sampling and DNA extraction in complex food, cosmetic, and pharmaceutical matrices to final signal analysis.

> Objective Decision Rules: Shifting away from arbitrary Cq cutoffs toward statistical decision rules tied directly to validation data near detection limits.

By establishing an international consensus through AOAC SEA Section and proposing a Molecular Biology Working Group for Halal Testing, this initiative aims to reduce analytical disputes, enhance interlaboratory comparability, and strengthen public trust in global halal supply chains.

Harmonizing Halal Analytical Methods: Key Insights from the 5th Annual Conference of the AOAC Southeast Asia Section

At the 5th Annual Conference of the AOAC Southeast Asia Section held today in Jakarta, Indonesia, Dwi Yulianto Laksono, S.Si., M.KPn. (Technical Director, SIG Laboratory), presented the findings of the global survey conducted by the Joint Expert Core Team on the Harmonization of Analytical Methods for Halal Food Certification.

Established on 02 April 2026, as a collaborative initiative between the AOAC Arab Section and AOAC Southeast Asia Section, the Joint Expert Core Team was formed to address global trade barriers, establish unified analytical guidelines, and build cross-regional collaboration.

Highlights from the Global Survey (June–July 2026):

> Broad International Participation: Gathered insights from 47 respondents spanning 14 countries across 6 regions—including testing laboratories, regulators, halal certification bodies, and global industry stakeholders.

> Addressing False-Negative Risks in Processed Matrices: While Real-Time PCR remains widely used, heavily processed matrices (e.g., gelatin) risk false-negative outcomes due to DNA degradation. Peptide-based techniques like LC-MS/MS and LC-HRMS offer greater stability and detection reliability. Qatar’s Ministry of Public Health (MOPH) successfully achieved a 0.1% Limit of Detection (LOD) for porcine markers using LC-MS/MS.

> Reference Standard Integrity Risks: The survey highlighted cases where commercial certified reference materials (CRMs) labeled as "Bovine" contained porcine markers upon LC-MS/MS testing, emphasizing the urgent need for verifiable standards.

> Operational Barriers Identified: High instrument investment costs, a lack of standardized sample preparation/digestion protocols, scarcity of traceable matrix-matched CRMs, and a shortage of specialized proteomic analysts remain key obstacles.

With pressing regulatory timelines—such as Indonesia’s upcoming Halal certification mandates—establishing unified, science-based benchmarks is essential for smooth international trade.
